In April 1995, (1) I presented my Ph.D. lecture on lipid peroxidation during the final year of my first clinical residency in Clinical Laboratory (1992-1995), which preceded my residency in Internal Medicine (1996-2001). Having trained and worked in both the US and UK, my experience underscores that medicine is a profession where learning never ceases. Today, I am pleased to recommend an article from the Nature Portfolio, published on May 10th, titled "Research progress on morphology and mechanism of programmed cell death," with a specific emphasis on ferroptosis. Could Digestive Enzymes Contribute to Aging? UCSD researchers have hypothesized a mechanism of aging called “autodigestion”- where digestive enzymes like trypsin, normally confined to the small intestine, leak into peripheral organs as we age. This leakage causes significant tissue damage, resulting in higher levels of collagen damage within organs linking this autodigestion to aging. Using Collagen Hybridizing Peptides (CHPs) to detect damaged collagen, the study revealed how this enzyme leakage accelerates aging and harms vital tissues. Here are their key findings:  Older rats showed higher collagen damage across multiple organs, including the liver, lungs, heart, brain, skin, and kidneys. This damage was closely linked to the presence of leaked digestive enzymes. When older rats were treated with a serine protease inhibitor for just two weeks, enzyme leakage and collagen damage were significantly reduced, offering a potential therapeutic approach. Implications: These findings imply that dietary habits, such as meal frequency and caloric intake, could influence the rate of enzymatic leakage and associated tissue damage, shedding a light on new avenues for therapeutic and lifestyle interventions to preserve tissue integrity and metabolic health. A neat approach for killing EBV-associated tumour cells. As with any genetic-based therapy though, a challenge will be delivering the nucleic acid to sufficient cells for a therapeutic benefit... Synthetic BZLF1-targeted transcriptional activator for efficient lytic induction therapy against EBV-associated epithelial cancers. Abstract The unique virus-cell interaction in Epstein-Barr virus (EBV)-associated malignancies implies targeting the viral latent-lytic switch is a promising therapeutic strategy. However, the lack of specific and efficient therapeutic agents to induce lytic cycle in these cancers is a major challenge facing clinical implementation. We develop a synthetic transcriptional activator that specifically activates endogenous BZLF1 and efficiently induces lytic reactivation in EBV-positive cancer cells. A lipid nanoparticle encapsulating nucleoside-modified mRNA which encodes a BZLF1-specific transcriptional activator (mTZ3-LNP) is synthesized for EBV-targeted therapy. Compared with conventional chemical inducers, mTZ3-LNP more efficiently activates EBV lytic gene expression in EBV-associated epithelial cancers. Here we show the potency and safety of treatment with mTZ3-LNP to suppress tumor growth in EBV-positive cancer models. The combination of mTZ3-LNP and ganciclovir yields highly selective cytotoxic effects of mRNA-based lytic induction therapy against EBV-positive tumor cells, indicating the potential of mRNA nanomedicine in the treatment of EBV-associated epithelial cancers. New Research Shows Promise of Llama-Derived Nanobodies for Targeting HIV A team of researchers led by Assistant Professor Jianliang Xu of Georgia State University has developed potent nanobodies derived from llama antibodies that show great potential for targeting and neutralizing hidden strains of HIV. The nanobodies are much smaller than conventional antibodies yet demonstrate remarkable effectiveness. When engineered into a "triple tandems" format by repeating small bits of DNA, the nanobodies were able to neutralize 96% of a diverse panel of HIV-1 strains in lab tests. Further analysis found the nanobodies mimic the recognition of the CD4 receptor, a key player in HIV infection. Fusing the nanobodies with broadly neutralizing antibodies resulted in molecules with unprecedented neutralizing abilities, potentially neutralizing close to 100% of strains. "Instead of developing a cocktail of antibodies, now we can make a single molecule that can neutralize HIV," said Prof. Xu. This research builds on over 15 years of work exploring llama antibodies for targeting HIV. Nanobodies derived from llamas' heavy-chain only antibodies are more flexible and effective than conventional antibodies. Prof. Xu is now collaborating with PhD candidate Payton Chan to further explore combinations of llama nanobodies and other broadly neutralizing antibodies towards developing new treatment options. Their goal is to achieve full neutralization of circulating HIV strains.
